The project aims at assessing the influence of different flow/mixing regimes on biomolecules crystallisation/nucleation process. Specific objectives:
-Implement different controlled flow and mixing regimes in microfluidic setups. Verification of the dedicated flows with a novel 3D particle image velocimetry technique.
-Screen for crystallisation conditions of the selected biomolecules applying counter diffusion methods.
-Develop a flow control methodology targeting a specific polymorph, and the required size distribution and morphology of the emerging crystals preventing the nucleation of protein aggregates.
-Implement an analytical optical approach, named Single Particle Extinction and Scattering (SPES) to classify the nucleated crystals/aggregates.
